Somehow (the problem occured right after doing my first "deploy") I have lost the vshost.exe.manifest to my project. That keeps me from rebuilding the application. Is there a way that I can recreate that file (I have searched for the file on my hard drive, including recycle bin and it is not present.)

I've never heard of anyone having problems rebuilding an app because the manifest file is missing. Are you building a C++ app In general the manifest file is auto-generated in that case. The VSHost binary is used for VS debugging so it is definitely not needed for deployment but the <app>.exe.manifest might be unless it is embedded in the app.

I believe that you are correct. My problem was elsewhere. It is true that I could no longer build my immediately after doing a deploy and I interpreted the error message that came up as indicating a missing file. I have worked around the problem by opening a new project and dragging all the relevant files from the original project into the new one. I can now rebuild the app, although I cannot see the design of my main form - instead I get the default empty form. Something is not pointing to something properly because I have another form in the project that does show the proper design.
